Fox Chase Cancer Center, Univ of Iowa, Univ of Pennsylvania. Utilizing the CEPH reference pedigree panel a a sample population, we have examined the linkage relationship of six loci previously localized to the short arm of chromosome 1. DNA polymorphisms for the glucose transport locus (GLUT), alkaline phosphatase locus (ALPL) and the pro natro dilation factor locus (PND) were used in combination with classical marker typing for PGD, PGM1, and RH available through the CEPH collaboration. Pairwise linkage analysis demonstrated 4 new significant linkages, RH-GLUT 90-0.24, z=4.51), RH-ALPL (0=0.10, z = 15.66), PND-RH (0=0.22, z=3.29) and FUCA1-ALPL (0=0.002, z=8.24). Detailed multipoint linkage analysis was performed on a 6 locus subset (PND, FUCA1, RH, GLUT, ALPL, PGM1). A preliminary order was first derived for the 6 loci by seriation. The likelihood of this order, and all other orders consistent with known physical localizations of the loci were evaluated. The best order was: PND - 17 - FUCA1 - 4 - RH - 7 - ALPL - 19 - GLUT - 29 Ð PGK1 where the interlocus intervals are percent recombination in males. The female rate was estimated to be 1.4 times the male rate.
