Ribonucleotide reductase (RR) catalyzes the formation of deoxyribonucleotides from corresponding ribonucleotides. It is composed of two non-identical subunits, proteins Ml and M2. RR activity is determined by M2 synthesis which is regulated in a cell cycle-dependent fashion. Chromosomal mapping of M2 sequences in human and mouse was carried out using a full-length mouse cDNA. Somatic cell hybrid analysis and in situ hybridization identified four sites of M2 related sequences in the human genome, located at 1p31-p33, lq21-q23, 2p24-p25 and Xp11-p21. In the mouse, M2 sequences were found on chromosomes 4, 7, 12 and 13 by somatic cell hybrid studies. In an attempt to identify the site of the active M2 locus, two other cDNA clones, p5-Beta and ornithine decarboxylase (ODC), which are co-amplified with RRM2 sequences in human and rodent hydroxyurea-resistant cell lines, were localized by Southern and in situ hybridization to human chromosome 2p24-p25, identified as one of M2 loci. Since this particular M2 sequence and p5-8 and ODC are most likely part of the same amplification unit, the RRM2 structural gene can be assigned to human chromosome 2p24-p25. This region is known to be homologous to a region of mouse chromosome 12 which likely carries the functional RRM2 locus in the mouse. By Southern analysis of human and mouse hydroxyurea resistant cells which overproduce M2, we have identified the amplified restriction fragments as those which were mapped to human chromosome 2 and to mouse chromosome 12.
