(A2006) C Szpirer, M Riviere, J Szpirer, M Genet, P Dreze, G Levan Universite Libre de Bruxelles, Department of Molecular Biology, B1640 Rhode-St-Genese, Belgium; University of Gothenburg, Department of Genetics S400 33 Gothenburg, Sweden (LG) Using cloned probes in Southern blot analysis of DNA extracted from a panel of l4 independent mouse hepatoma x rat hepatocyte hybrids which segregate rat chromosomes, we determined the localization of the following 10 rat loci: alpha1-glycoprotein (orosomucoid; ORM), alpha2u-globulin (major urinary protein; MUP) atrial natriuretic factor (ANF = PND), glycoprotein-4-Beta-galactosyltransferase (GGTB), interferon alpha (IFNA, several genes), interferon Beta (IFNB), LCK and MOS oncogenes, D4Rp1 and a region of proviral insertion in MoMuLV-induced rat thymomas (DS1-1). All these loci cosegregated with rat chromosome 5 (at least 3 discordant clones were obtained for all the other chromosomes). Previously, we also assigned the MYCL oncogene and the aldolase B (ALDOB) gene to rat chromosome 5. These data show that rat chromosome 5 is highly homologous to mouse chromosome 4, known to carry most of these genes (Orm, Mup, Pnd, Ggtb, Ifa, Ifb, Lck, Mos, Dsi-1, D4Rp1, Mycl), and shares several loci with human chromosomes 9 (9q for ORM and 9p for the IFNA, IFNB and GGTB genes), or with the short arm of human chromosome 1 (1p carries PND, LCK and MYCL). In addition, using somatic cell hybrids possessing a single deleted rat chromosome 5, the rat IFN genes could be regionally localized to the region 5q22-23, which has also been shown to contain a locus suppressing the transformed phenotype (Islam et al., J. Cell Sci., in press); all the other loci maps outside region 5q22-23. On the other hand, we also found that the mouse Lck probe detects two non-syntenic genes in the rat genome: one maps on chromosome 5 (see above) and the other on chromosome 7 (which is partially homologous to mouse chromosome 15).
