abstractText | Full text of MNL contribution: Mapping of mdx - A correction. Our laboratory previously reported an X chromosome gene order of Hprt, mdx, Cf-8/Rsvp, Pgk-1 which was based upon the analysis of backcross male progeny that had been previously typed for segregation at the Hprt, mdx and Pgk-1 genes. We specifically analyzed a subset of males that were recombinant between Hprt and Pgk-1 for segregation of Cf-8 and Rsvp. Those analyses were based upon results which indicated that the parents of the backcross were informative heterozygotes for Cf-8 and Rsvp. We have subsequently discovered that one of the female progenitors of the backcross series was not heterozygous at those two loci even though she was informative for Hprt, mdx and Pgk-1. Thus, several backcross males from this female parent gave phenotypic scores for Cf-8 and Rsvp that were overtly recombinant between mdx and Pgk-1. We have reanalyzed the backcross progeny without this female lineage which eliminated the recombinant individuals that gave our previously reported gene order. In subsequent tests we have analyzed seven additional backcross males that were Hprt - Pgk-1 recombinants. The female parents of these males was heterozygous for Rsvp and Cf-8 and these backcross males were test mated to verify transmission of the mdx genotype. One male was recombinant between Cf-8/Rsvp and mdx and produced test cross progeny which verified his genotype as Hprt-a, Rsvp-c, mdx, Pgk-1b (where Rsvp-c defines the non-C57BL/lOSn.mdx allele). Thus, a gene order of Hprt, Cf-8, Rsvp, mdx and Pgk-1 is consistent with these results. This map order is consistent with that reported for C57BL/lOSn.mdx x g. spretus crosses reported by other laboratories. (V.M. Chapman, D.A. Stephenson, L.J. Mullins and S.G. Grant) |
