abstractText | Full text of Mouse Genome contribution: 3. New multiple recessive linkage testing stock. Our laboratory recently described a new linkage testing stock (MEV/1Ty-Hm Sl CaJ) homozygous for 11 ecotropic murine leukemia virus proviruses and segregating three dominant visibles (Taylor and Rowe, Genomics 5:221-32). This stock suffices to 'sweep' about half of the autosomal genome. We have now constructed a new multiple recessive stock designed to complement the MEV stock. The new stock carries nonagouti (a; Chr 2), brown (b; Chr 4), waved-1 (wa-1; Chr 6), beige-8J (bg8J; Chr 13), and piebald (s; Chr 14). The stock gets b, and wa-1 from ABP/Le, bg8J from DBA/2J-bg8J, and s from C3HeB/FeJ-a s. From the F2 generation of ABP/Le (a/a b/b wa-1/wa-1 p/p se/se bt/bt) crossed to DBA/2J-bg8J (a/a b/b d/d bg8J/bg8J), mice homozygous for a, b, wa-1, se, and bg8J were selected while brother-sister mating. Once this stock was established, it was crossed to C3HeB/FeJ-a s, and these F1s were backcrossed to the a, b, wa-1, se, bg8J stock. Pairs of these N2 mice, homozygous for various combinations of a, b, wa-1, and bg8J, were mated inter se. The piebald offspring produced by some of these pairs were then sib-mated for several generations until mice homozygous for a, b, wa-1, bg8J, and s (but apparently not p, se or bt) were obtained. Mice from two such sublines were crossed to establish the five recessive stock. This stock, which is being bred to avoid close inbreeding, shows good viability and fertility. We propose to call the stock MRT (multiple recessive stock, Taylor). It is estimated that the autosomal contributions of the three ancestral strains (ABP/Le, DBA/2J, and C3HeB/FeJ) are 37.5%, 37.5%, and 25%, respectively. At this point, there is a possibility that pink-eyed dilution (p), short-ear (se), and/or belted (bt) may also be carried heterozygously by some individuals. By screening the equivalent of 50 fully informative gametes from a cross involving MRT, it is possible to sweep an additional 25% of the genome not screened by the MEV markers.
