| abstractText | Full text of MNL contribution: Research News: Linkage of Tpi-1 and Gapd. In the offspring of paternally treated mice, mutants with approximately 50% reduction in triosephosphate isomerase (TPI) or glyceraldhyde-3-phosphate dehydrogenase (GAPDH) activity have been detected (Mut. Res. 160 [1986] 243-248, 176 [1987] 81-91). Peters and Bulfield have determined the position of Tpi-1 on chromosome 6 (MNL 70 [1984] 82). Mouse GAPDH has been shown to be syntenic with chromosome 6 and with Tpi-1 in a series of mouse x Chinese hamster cell hybrids (Cytogenet. Cell. Genet. 25 [1979] 139). We have used one of our TPI mutants (#9606) and 4 GAPDH mutants (#525, #577, #3284 and #28003) to determine the genetic distance between Tpi-1 and Gapd. For linkage testing, crosses were made of double heterozygotes in repulsion (Tpi-1 +/+ Gapd) and wild types (+ +/+ +). The results are as follows: GAPDH mutant: #525; Offspring phenotype: Tpi-1 +: 62; + Gapd: 69; Tpi-1 Gapd: 0; + +: 0. GAPDH mutant: #577; Offspring phenotype: Tpi-1 +: 124; + Gapd: 93; Tpi-1 Gapd: 0; + +: 1. GAPDH mutant: #3284; Offspring phenotype: Tpi-1 +: 73; + Gapd: 82; Tpi-1 Gapd: 0; + +: 0. GAPDH mutant: #28003; Offspring phenotype: Tpi-1 +: 80; + Gapd: 69; Tpi-1 Gapd: 0; + +: 0. Total: Tpi-1 +: 339; + Gapd: 313; Tpi-1 Gapd: 0; + +: 1. The recombination frequency for the pooled data is 0.15% (SD = 0.15%) implying a very close linkage of Tpi-1 and Gapd. (W. Pretsch) |
