molecularNote | The translation of the immunoreceptor tyrosine-based activation motif (ITAM) was precluded by the generation of a nonsense mutation at codon 181 (exon 4). Sequence analysis revealed that the incorporation of a targeting vector containing a floxed neo cassette downstream of the endogenous locus and tyrosine to phenylalanine missense mutations in the ITAM resulted in a 5 nt deletion (4506 to 4510) thereby generating the stop codon at position 181. The neo cassette was excised in vitro via cre mediated recombination. The resultant protein was expected to lack 40 carboxy terminal amino acids, and thus retain B cell surface Ig expression but lack endogenous signaling. |
