| molecularNote | A targeting vector was designed to remove a 6 kb region containing the antisense control region, expecting that the transcription of the antisense RNA would be disrupted without altering the protein coding sequence of the sense RNA. The ASC was replaced with a neomycin resistance cassette. In situ hybridization of mutant E16.5 brains revealed that expression of Zfhx2os was under the detection limit, whereas that of Zfhx2 was increased in various areas of the brain, including ectopic expression in the pontine nuclei, where wild-type expresses Zfhx2os and not Zfhx2. Northern blot detected transcript from Zfhx2 but not Zfhx2os. |
