| molecularNote | Two loxP sites were inserted flanking exon 3. Preceding the floxed exon 3, a reporter/selection cassette (SA-mCFP-Neo) flanked by two Frt sites, containing a splice acceptor, a membrane-targeted cyan fluorescent protein reporter gene and a neo cassette, was inserted. After Cre-mediated excision of exon 3, a frameshift in the downstream coding sequence resulted in a null allele and a functional mCFP reporter. Western blot analysis on total brain extracts and immunostaining on adult brain tissue confirmed the absence of the protein in homozygous mutants. Immunohistochemistry also confirmed mCFP expression in the hippocampus. |
