molecularNote | The "RasE Multi-Hit BAC" (bacterial artificial chromosome) includes three independent expression modules, each containing the CAGGS promoter (comprising the cytomegalovirus immediate early (CMV-IE) enhancer and the chicken beta-actin promoter, first exon and first intron, with the 3' splice junction from the rabbit beta-globin gene); a mutant Hras gene combining the G12V (glycine to valine at amino acid (aa) 12) mutation with either T35S (threonine to serine at aa 35) (HrasV12S35), E37G (glutamate to glycine at aa 37) (HrasV12G37), or Y40C (tyrosine to cysteine at aa 40) (HrasV12C40); an internal ribosomal entry site; a fluorescent reporter gene (eYFP, dsRed, or hCD2t, respectively); and an SV40 polyadenylation signal; each module flanked by chicken 2xHS4 insulator sequences. This construct was recombined into the PstI restriction site of a BAC containing the mouse collagen, type I, alpha 1 (Col1a1) locus. Transgenic mouse line 290 was shown by Southern blot analysis to have integrated a single copy of the BAC. |
