| molecularNote | The L1L2_Bact_P cassette was inserted at position 145877259 of Chromosome 3 upstream of the critical exon (exon 4) (Build GRCm39). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. Insertion of this cassette creates a reporter knockout mouse. Cre expression removed the neomycin selection cassette. Western blot analysis using rabbit anti-MCOLN2 antibody confirmed the absence of protein in bone marrow-derived macrophages (BMDMs) isolated from homozygous mutant mice both under resting conditions and upon BMDM activation with LPS. |
