molecularNote | CAGâroxâStopâroxâConfetti was targeted to the Rosa26 gene locus with four fluorescent protein sequences uniquely positioned in a tandem fashion and delimited by loxP sites in the opposite orientations. Specifically, it is composed of two adjacent floxed head-to-tail tandem dimers. The first head-to-tail dimer contains a loxP site and mCFP in forward orientation and RFP in reverse orientation followed by a reverese loxP site. The second head-to-tail dimer contains a loxP site and YFP in forward orientation, and nGFP in reverse orientation followed by another reverse loxP site. The allele contains roxâStopârox cassettes before confetti, so that only after dual Dreârox and CreâloxP recombination could single-color fluorescence reporters such as yellow fluorescent protein (YFP), green fluorescent protein (GFP) and red fluorescent protein (RFP) be expressed in Dre+Cre+ cells and their descendants. Without cre recombination, only mCerulean CFP is expressed after Dre-rox recombination. |
