| molecularNote | The targeting vector was designed to insert a Cre/ERT2 fusion protein into the tenascin C (Tnc) locus. Specifically, this vector includes (from 5' to 3') an FRT-flanked Neomycin-STOP, a cre/ERT2 fusion gene (Cre recombinase fused to a human estrogen receptor ligand binding domain) and rabbit beta-globin gene polyadenylation signal. The construct was electroporated into (C57BL/6NTac x 129S6/SvEvTac)F1-derived iTL BA1 embryonic stem (ES) cells and correctly targeted ES cells were injected into recipient blastocysts. The resulting chimeric mice were identified by genotyping and bred to a C57BL/6 FLP mouse line to remove the neomycin cassette. |
